The 15-year-old disappeared near her home in Castlederg after she went to a disco in Bundoran, Co Donegal with friends.
Extensive searches were carried out but no firm evidence of the teenager being alive or dead have ever been found.
Convicted child killer Robert Howard was cleared of her murder in 2005 after his past convictions for rape, murder and child abuse were withheld from the jury.
Howard is serving life for the murder of 14-year-old Hannah Williams, whose body was found in a cement works in Kent in 2002.
Police have not disclosed what developments have led to the investigation, but officers previously said they would only conduct a new search for Arlene's body if fresh evidence came to light.
A report by the Police Ombudsman in 2008 said police treated Arlene`s disappearance as a missing person inquiry for too long rather than a potential abduction or murder.
It also said officers took too long to arrest main suspect Howard.
Although they had grounds to do this within 48 hours of her disappearance, it did not happen until 46 days after Arlene was reported missing.
As a result, police were not able to get the clothing Howard wore on the night of the disappearance and it may have allowed him the opportunity to dispose of evidence.
